A metagenomic view of novel microbial and metabolic diversity found within the deep terrestrial biosphere at DeMMO: A microbial observatory in South Dakota, USADepartment of Earth and Planetary Sciences, Northwestern University, Evanston, Illinois, USAMagdalena R. Osburn, Department of Earth, Environmental, and Planetary Sciences, Northwestern University, 2145 Sheridan Road, Technological Institute F379, Evanston IL, 60208, USA.

In this study, we use whole metagenome sequencing methods to investigate microbes from the Deep Mine Microbial Observatory located in the Sanford Underground Research Facility in Lead, South Dakota, USA. We draw on years of geochemical characterization to integrate microbial, geochemical, and geological observations to expand our genomic interpretation to subsurface microbial processes.

Of the genomes assembled here, perhaps some of the most interesting are the near complete, or ‘complete’, members of the newly identified Goldbacteria , Edwardsbacteria , and Eisenbacteria .

Members of the phylum Omnitrophota are particularly well represented in our dataset with MAGs originating from across DeMMO sites. The phylum was originally dubbed ‘Omnitrophica’ in 2013 and has received much attention in recent years, as they are a widespread and abundant candidate phylum in global natural environments such as sediment, non-saline ground and surface water, and soils .
