Eleven's fate, new villains and #Justice4Barb.

The second season begins in October 1984, as we see a teenage girl with psychic powers and the number 008 tattooed on her arm rob a bank in Pittsburgh.Her arrival isn’t the only change in Hawkins. Joyce is dating her former school classmate Bob , while her son Will has been experiencing visions of the Upside Down and a shadowy monster.

While he continues to keep her hidden, much to Eleven’s frustration, Hopper investigates a substance in fields where pumpkins are rotting and confronts Owens about it. Nancy and Jonathan, meanwhile, are attempting to contact Barb’s mother to tell her the truth about what happened to her daughter, but they are intercepted and taken to the lab, where Owens explains that Barb died in the Upside Down, and he wants to keep the place a secret from the world. .

While all this is going on, Nancy and Jonathan have shared Owen’s confession with Murray, and the trio go on to send a watered down version to newspapers around the country. Lucas and Max – who have been spending time together when Max’s stepbrother Billy doesn’t get in the way – team up with Dustin and Steve and attempt to lure Dart to a junkyard, only for the creature to turn up with a pack of gruesome friends. The monsters run away, however, and attack Owen’s team on their way to the lab.

Mike, Joyce, Bob, Hopper and Owens take Will to the security room, but when the power goes down, Bob volunteers to go and reset the system, giving the others time to escape. Unfortunately, while the others get away, Bob – poor, sweet, selfless Bob – is trapped and eaten by the creatures in front of an understandably distressed Joyce.

Single-cell multi-omics profiling links dynamic DNA methylation to cell fate decisions during mouse early organogenesis - Genome BiologyBackground Perturbation of DNA methyltransferases (DNMTs) and of the active DNA demethylation pathway via ten-eleven translocation (TET) methylcytosine dioxygenases results in severe developmental defects and embryonic lethality. Dynamic control of DNA methylation is therefore vital for embryogenesis, yet the underlying mechanisms remain poorly understood. Results Here we report a single-cell transcriptomic atlas from Dnmt and Tet mutant mouse embryos during early organogenesis. We show that both the maintenance and de novo methyltransferase enzymes are dispensable for the formation of all major cell types at E8.5. However, DNA methyltransferases are required for silencing of prior or alternative cell fates such as pluripotency and extraembryonic programmes. Deletion of all three TET enzymes produces substantial lineage biases, in particular, a failure to generate primitive erythrocytes. Single-cell multi-omics profiling moreover reveals that this is linked to a failure to demethylate distal regulatory elements in Tet triple-knockout embryos. Conclusions This study provides a detailed analysis of the effects of perturbing DNA methylation on mouse organogenesis at a whole organism scale and affords new insights into the regulatory mechanisms of cell fate decisions.
