Enzyme drives cognitive decline in mice, provides new target for Alzheimer's ucsandiego NatureComms

. Connections were limited to high confidence with a maximum of 20 connections for the second shell. First shell of interactions was restricted to the query only.Mice were deeply anesthetized with ketamine, and perfused with 0.9% w/v Sodium Chloride, then perfused with 4% paraformaldehyde in phosphate buffer . The brain tissues were removed and post-fixed in 4% PFA in PB for 30 min. Using a vibratome, 100 µm coronal sections were sliced and stored in 1× dPBS.

Antibodies: anti-PKCα antibody was from BD Transduction Laboratories. β-Actin antibody was purchased from Sigma-Aldrich , total SAP97 was from Enzo Life Sciences , phospho-MARCKS and total MARCKS were obtained from Santa Cruz Biotechnology . GAPDH , vinculin , phospho- PKC substrate , phospho-ERK1/2 and total ERK1/2 antibodies were purchased from Cell Signaling Technology, and were used at a 1:1000 dilution.
